The whole macronutrient isolation is a theory that's been popularized (to my knowledge) by a nutritionist named John Berardi. The idea is that you don't want a lot of triglycerides and a lot of insulin moving around in your system at the same time. While it certainly makes sense, fat is the last macro to be metabolized as energy since it is easiest to store, I don't think that it will make a huge difference in terms of effects on your physique (provided you're not downing fettucine alfredo all the time). The theory is that since fat is at the bottom os the list to be metabolized, that your body will use the carbs and immediately shuttle the triglycerides to adipose tissue. Like I mentioned, it seems to make sense. Berardi recommends separating your meals into protein-fat meals and protein-carb meals. At your P+C meals, you don't want to ingest more than 5-10g of fat and with the P+F meals, you want to keep your carbs to less than 10-15g, I believe. You're supposed to consume P+C meals post training for half your meals and the P+F meals at other times. That theory is fine for those that train in the morning, but your don't want to be ingesting a lot of carbs in the evening. I follow the rules to some degree, eating P+C meals in the morning and P+F meals in the evening and allowing myself to mix macros more (talk about allitertation) during the day. I try to make coming home from work my carb cut-off. I think that formula is a little easier to follow. Of course you want your post training meal to still be P+C, so on training days (I train after work) I'm forced to break the rules a tad.
I think ultimately the macro isolation strategy will make a difference, but it won't be dramatic.

A diet higher in protein/fat ratio can be beneficial at certain times. In fact, a 50/30/20, P/F/C ratio is an excellent bodybuilding diet for a number of reasons. After getting passed the initial vilification of fat from the media, think this. If you have gained muscle for instance, and want to hold it for a period of time, or trying to gain muscle, this can be an excellent way. We all know what happens when excess carbs are consumed and glycogen stores are full, and you exceed your maintenance calories. You are trying to increase muscle tissue, not just add water to the cells with sugar and creatine. So to avoid this, healthy fats can substitute for the carbs. Without going into the great benefits of healthy fats here, just know it. They are very calorie dense for one, 9 cal. per gr. as compared to 4 for carbs, which provide the calories you need. This along with protein will provide the number of calories you need to take advantage of training.

Yeah, noirutla, he pushes Biotest supps HARD. I currently use Grow!, but that's really it. He also pushes things like needing 2-2.5x your weight (in lbs.!) in protein that really makes me doubt his intentions. Although I think Berardi knows his stuff, I think he's more committed to Biotest than his readers.
