What specific muscle groups are isolated by laying out a tractor tire and smacking the crap out of it with a sledge hammer?

That's a terrible question, since the idea of training like this is not to "isolate" any muscle group, but rather teach the body to act as a whole.
However, although exercises like this are most often used for GPP and total body conditioning, you could consider this an anterior chain exercise.

Like wrestler said it's a total body conditioning. It's often used with escalating density training.

I've only once seen this kind of workout being promoted on some site that was selling training hammers. The hammers were extremely expensive and the site seemed kind of shifty...still though, I'd love to get my hands on a 100lb. sledge.

Tire+sledge shouldnt be too expensive. You can probably find a tractor tire at a junkyard and sledges usually dont cost that much...
this is a very good conditioning workout....it's used by a few fighters....most notably fedor ..

You shouldn't ever pay for a tire. Most places are happy to get rid of them.
My buddy Jared actually carries around 2 sledge hammers in the back of his Civic at all times. Kind of weird, but w/e.
