I was 6'1 and 298 when I started! For a beginner, it's important not to go crazy out from the start. The best routines are the ones that a person can realistically follow.
For starters get a free strength training routine from this site. Once you are done lifting, try hitting the treadmill for some very light cardio for 10-20 minutes. Then on your "off days" from lifting, try a more intense cardio session of 20-40 minutes. My schedule is Mon,Wed,Fri,for lifting and Tue,Thur for real cardio.
Many people think that cardio has to be running, but the truth is that anything you do burns calories. So don't kill yourself for net being a track star from the get go.
You'll also need to get your diet pinned down because it's the #1 factor in any continued success.

Hi paradigm and welcome, when i started my home training, about 10 weeks ago, i was working out on Mon, Wed and Fri, on Tue and Thur i started to walk round the block a couple of times, the more i did my home training, the fitter i got, the fitter i got, the further i could walk, now i walk between 4 and 5 miles twice a week, so like returnofplex says, start with what you feel comfortable with and build up over the weeks, pushing yourself further each week.
Good luck, eat well, train well and keep us all posted with your progress.
